Definition of ZD9331

An orally available non-polyglutamatable quinazoline derivative with potential antineoplastic activity. Plevitrexed, behaves as a folate analogue, is transported into the cell via alpha isoform of the folate receptor (alpha-FR) and, to a lesser extent, by reduced folate carrier (RFC). Intracellularly, this agent selectively binds to the folate binding site of thymidylate synthase, thereby hampering thymidine synthesis that leads to dna synthesis inhibition and subsequently inducing apoptosis. RFC is ubiquitously expressed in normal tissue, alpha-FR, however, is overexpressed in a variety of tumors while its expression in normal tissue is very low.
